ANTİDEPRESAN İLAÇLAR VE GENOTOKSİSİTE

Abstract

Depresyon sinirlilik, uykusuzluk, yorgunluk, ajitasyon, psikomotor değişiklikler, suçluluk, kendine değer vermeme ile karakterize kompleks bir hastalıktır. Bu hastalığın tedavisinde en sık kullanılan yöntem antidepresan kullanımıdır.  Antidepresanlar çok sayıda hastada uzun süreli kullanılan ilaçlardır ve literatürde bunların genotoksik etkilerini rapor eden çeşitli çalışmalar vardır. İlaçlar piyasaya sürülmeden önce genotoksisite testlerini de içeren bazı testlerin yapılması gereklidir. Genotoksisite çalışmaları bir kimyasalın DNA hasarına neden olup olmadığını belirler. Bu DNA hasarı tek ve çift zincir kırıkları, eksizyon onarım kaybı, çapraz bağlar, alkali labil bölgeler, nokta mutasyonları ve yapısal ve sayısal kromozomal anomaliler şeklinde olabilir. Günümüzde, antidepresan kullanımının hızla artması sonucunda bu ilaçların genetik yapıda olumsuz etkileri olup olmadığının tespit edilmesi son derece önem kazanmıştır. Bakterilerde, in vivo ve in vitro memeli hücrelerinde, bitkilerde veya Drosophila’da çeşitli test sistemleri herhangi bir kimyasal ajanın indüklediği genetik hasarı değerlendirmek için kullanılmaktadır. Bu çalışmada, farklı test sistemlerinde antidepresanların daha önce yayınlanmış genotoksisite test sonuçları derlenmiştir.
