trnL İntron ve trnL-F Bölgeleri ile Van İlinde Bulunan Orkide (Orchidaceae) Türlerinin Filogenetik İlişkisinin Belirlenmesi

Öz

Van ilinde doğal olarak yayılış gösteren Orkide (Orchis) cinsine ait dokuz türün filogenetik ilişkisi kloroplast DNA’sında bulunan trnL intron ve trnL-F IGS bölgeleri kullanılarak belirlenmeye çalışılmıştır. Aynı cinse ait diğer türlerle olan evrimsel ilişkileri de göstermek için farklı türlerin DNA sekansları NCBI veri tabanından alınarak analize dahil edilmiştir. Her bir tür için ikili okuma yapılarak en doğru DNA sekansı alınmış ve bunlar hizalanarak Maximum Likelihood (ML) methodu ile filogenetik ağaçlar çizdirilmiştir. trnL-F bölgesinde varyasyon çok daha az olmasına rağmen elde edilen ağaç topolojileri oldukça benzerdir. Heriki ağaçta da altcinsler (Orchis, Anacamptis ve Neotinea) farklı kladlarda yer almış ve alt seksiyon seviyesinde tür ayrımı yapılabilmiştir. Çalışma sonunda Anacamptis ve Neotinea altcinslerinde bulunan türlerin taksonomik pozisyonunun değiştirilerek cins seviyesine çıkarılması gerektiği sonucuna varılmıştır.
