Leaf and Peduncle Anatomy of Species Belonging to Hymenostegis Bunge and Hymenocoleus Bunge Sections of Genus Astragalus L. (Fabaceae) in Türkiye

Abstract

The aim of this study is to examine and compare the peduncle and leaf anatomy of the taxa Hymenostegis Bunge and Hymenocoleus Bunge of Astragalus sections under a light microscope. This study aims to synthesize new data obtained from the peduncle and leaf anatomy of the species with morphological characters. Thus, it is intended to enrich the key characters used in species discrimination and provide an original anatomical contribution to resolving the systematic problems of the relevant taxa. The collected specimens were sectioned using a microtome and stained with safranin and fast green. Photographs were taken under a light microscope. A total of 56 anatomical characters (both qualitative and quantitative) were analyzed, including cell dimensions from epidermis, collenchyma, parenchyma, sclerenchyma, phloem, xylem, and pith tissues. Cell diameter measurements were made for these quantitative characters. A phylogenetic tree (UPGMA) was constructed to demonstrate the relationships among taxa. The peduncle, rachis, and leaflet anatomy of 13 species of the genus Astragalus exhibited marked differentiation among species and sections. A. vaginans (sect. Hymenocoleus) consistently exhibited the largest dimensions across all tissues, clearly distinguishing it from Hymenostegis species. Conversely, A. zohrabi represented the lower limit of anatomical variation. Multivariate analyses (UPGMA and PCoA) confirmed the isolated position of A. vaginans, while Hymenostegis species formed concordant subclusters. In general, vascular bundle characteristics, sclerenchyma thickness, and pith development were identified as the most diagnostic anatomical characters for distinguishing the studied taxa. The marked anatomical differences observed among the studied Astragalus species clearly distinguish A. vaginans from members of the Hymenostegis section. Its larger cell dimensions, broader pith region, and thicker vascular and sclerenchymatic tissues, together with its isolated position in UPGMA and PCoA analyses, strongly support its sectional separation within Hymenocoleus. In contrast, A. zohrabi represents the lower limit of anatomical variation. The congruence between anatomical differentiation, multivariate clustering, and geographical proximity suggests that regional ecological conditions have played an important role in shaping evolutionary divergence within this group. As a result, our findings demonstrate that internal anatomical characters provide reliable indicators for species discrimination within Astragalus and that leaf and peduncle anatomy have taxonomic value in distinguishing species.

Ethical Statement

The research does not include any experimental application, intervention or clinical procedure on humans or experimental animals. Only plant samples and herbarium materials collected from nature within the framework of legal permissions were used within the scope of the study. Since there is no use of live animals, genetic intervention or any application based on human data, it does not fall within the scope of a research requiring ethics committee approval.
