Isolation and Identification of Antagonistic and Plant Growth-Promoting Endophytic Bacteria for the Management of Pectobacterium betavasculorum in Sugar Beet

Abstract

This study aimed to isolate and identify endophytic bacteria from sugar beet (Beta vulgaris L.) grown in Yozgat province and to evaluate their biological control and plant growth-promoting (PGPB) potential against Pectobacterium betavasculorum, the causal agent of bacterial vascular necrosis and root rot. A total of 120 sugar beet tuber samples were collected, yielding 325 bacterial isolates. After preliminary screening, 221 isolates were retained. Pectolytic activity on potato, hypersensitive reaction on tobacco, and growth at 37 °C were tested to exclude potentially pathogenic isolates. Ultimately, 55 endophytic isolates were evaluated for antagonistic activity and PGP traits, including protease production, phosphate solubilization, indole-3-acetic acid (IAA) production, and ammonia production. Promising isolates were identified by MALDI-TOF MS. Among the isolates, 7 showed antagonistic activity against plant pathogens, 24 produced protease, 7 solubilized phosphate, 19 produced IAA, and 30 produced ammonia. Twelve promising isolates were identified, including Raoultella ornithinolytica, Bacillus thuringiensis israelensis, Paenibacillus polymyxa, and Serratia rubidaea. S. rubidaea SB 65.2 demonstrated the highest multifunctional potential. The results indicate that sugar beet harbors diverse endophytic bacteria with multiple biocontrol and PGP mechanisms, highlighting their ecological and functional diversity. Endophytic bacteria isolated from sugar beet represent promising candidates for sustainable management of P. betavasculorum and may contribute to environmentally friendly agricultural practices.
