Molecular Detection of Infectious Viruses in Potato Tubers in Potato Production Areas in Tokat Province

Abstract

Ninety-one potato tuber samples showing virus symptoms were collected from potato production areas in Niksar, Erbaa, and Artova districts, where potato cultivation is done in Tokat province. Tuber samples were subjected to RT-PCR tests to determine the presence of potato virus Y (PVY), potato virus S (PVS), potato virus X (PVX), and potato leafroll virus (PLRV) viruses. Single or mixed virus infections were detected in 68 (74.72%) of the tuber samples tested. Of the 91 germinated tubers, 67 were infected with PVY (73.62%), 6 were infected with PVS (6.59%), and one was infected with PLRV (1.09%). In addition, PVY+PVS mixed infection was detected in 5 of the tubers infected with PVS, while PVY infection was detected in the tubers infected with PLRV. It was determined that the most common virus in tuber samples collected from different districts of Tokat province was PVY, followed by PVS. Infected tubers used as seeds cause serious losses in yield from year to year. The use of virus-free and certified seeds is the most important factor in preventing viral-related losses. otato leafroll virus (PLRV) viruses. Tokat İli Patates Üretim Alanlarında Patates Yumrularında Enfeksiyon Oluşturan Virüslerin Moleküler Olarak Tespiti

Abstract

Tokat ilinde patates yetiştiriciliği yapılan Niksar, Erbaa ve Artova ilçelerindeki patates üretim alanlarından virüs belirtisi gösteren patates bitkilerinden 91 adet yumru örneği toplanmıştır. Yumru örnekleri, potato virus Y (PVY), potato virus S (PVS), potato virus X (PVX) ve potato leafroll virus (PLRV) etmenlerinin varlığını tespit etmek amacıyla RT-PCR testine tabi tutulmuştur. Test edilen yumru örneklerinin 68 tanesinde (%74.72) tekli veya karışık virüs enfeksiyonun varlığı tespit edilmiştir. Çimlendirilen 91 yumrunun 67’si PVY (%73.62), 6’sı PVS (%6.59) ve 1 tanesi de PLRV (%1.09) ile enfekteli bulunmuştur. Ayrıca PVS ile enfekteli bulunan yumrulardan 5 tanesinde PVY+PVS karışık enfeksiyonu belirlenirken, PLRV ile enfekteli yumruda da PVY enfeksiyonu tespit edilmiştir. Tokat ilinde farklı ilçelerden toplanan yumrularda en yaygın PVY görülürken bunu PVS’ etmeninin izlediği belirlenmiştir. Tohumluk olarak kullanılan enfekteli yumrular yıldan yıla verimde ciddi kayıplara neden olmaktadır. Virüsten ari ve sertifikalı tohumlukların kullanılması viral kaynaklı kaybın önüne geçilmesinde en önemli faktördür.
